Inhibition of miR-3148 could promote myocardial mitophagy and reduce apoptosis in AMI mice. (a and b) WB analysis was performed to examine the expression levels of PINK1, Parkin, Beclin1, p62, LC3 I and LC3 II proteins associated with mitophagy. (c) The PINK1 and Parkin expression were measured by qRT-PCR. (d and e) Levels of apoptosis-related proteins, including Caspase 9, Caspase 3, and Cyt C, were evaluated through WB analysis. (f) TUNEL staining of mice myocardium. Apoptotic cells that were TUNEL-positive appear brown, with hematoxylin staining revealing blue nuclei. Cells that were TUNEL-positive typically cannot be further stained with hematoxylin. Bar = 50 μm. (g) Comparison of the number of TUNEL positive cells of mice among groups.
