Overview

PPP2R1B encodes the beta isoform of the regulatory subunit A (also known as PR65 or scaffold subunit) of protein phosphatase 2A (PP2A), a major serine/threonine phosphatase critical for tau dephosphorylation in the brain[@liu2005]. PP2A dysfunction is centrally implicated in Alzheimer's disease (AD) and related tauopathies, where reduced phosphatase activity leads to hyperphosphorylation of the microtubule-associated protein tau and subsequent neurofibrillary tangle formation[@gong2006]. The PP2A holoenzyme is a heterotrimeric complex comprising a catalytic subunit (PP2A-C), a structural subunit (PP2A-A, encoded by PPP2R1A or PPP2R1B), and one of multiple regulatory subunits (PP2A-B) that together determine substrate specificity, subcellular localization, and catalytic activity[@xing2006].
