OGDHL (Oxoglutarate Dehydrogenase Like) is a gene encoding a protein with high similarity to the E1 component of the alpha-ketoglutarate dehydrogenase complex (alpha-KGDH) in the tricarboxylic acid (TCA) cycle["@sheeran2018"]. While OGDHL lacks the full enzymatic activity of canonical alpha-KGDH, it plays important roles in mitochondrial metabolism, oxidative stress response, and neuronal function["@bhardwaj2020"]. OGDHL is predominantly expressed in brain tissue, particularly in [neurons](/entities/neurons) with high metabolic demands.
Function
OGDHL exhibits several key functions:
Metabolic Regulation: Modulates mitochondrial TCA cycle flux and energy production[@gibson2020]
Oxidative Stress Response: Influences cellular redox status through α-ketoglutarate metabolism
Neuroprotective Properties: Protects neurons against oxidative stress and metabolic dysfunction[@park2019]
Epigenetic Regulation: Provides α-ketoglutarate as a cofactor for DNA demethylases (TET enzymes)[@tahiliani2009]
Disease Associations
Alzheimer's Disease
OGDHL expression is significantly reduced in AD brains, particularly in the [hippocampus](/brain-regions/hippocampus) and frontal [cortex](/brain-regions/cortex)[@bubber2005]. This downregulation correlates with decreased TCA cycle activity and impaired glucose metabolism, both hallmark features of AD. OGDHL deficiency may contribute to bioenergetic failure in AD neurons[@yin2019].
Parkinson's Disease
In PD, OGDHL expression is altered in the substantia nigra[@elstner2011]. The enzyme's role in mitochondrial function makes it a relevant player in PD pathogenesis, where mitochondrial dysfunction is a central mechanism.
Epilepsy
OGDHL variants have been associated with epilepsy phenotypes, reflecting its importance in neuronal energy metabolism[@liu2018].
