Exploratory experiment designed to discover new patterns targeting HDAC4, NHE6, LAMP2 in Microglia and astrocytes. Primary outcome: Enhanced Aβ clearance through neuroimmune modulation
This experiment examined how the HDAC4-NHE6-pH axis modulates the neuroimmune microenvironment to enhance amyloid-β clearance through multiple synergistic mechanisms. The study investigated upregulation of phagocytic receptors and microglial recruitment for Aβ plaque phagocytosis. Additionally, it examined reactivation of autophagy-lysosomal function in astrocytes through increased LAMP2 expression. This comprehensive analysis demonstrated how pH regulation affects both microglial and astrocytic functions in Aβ clearance, providing evidence for the multifaceted therapeutic approach of targeting the HDAC4-NHE6 pathway.
Assessment of phagocytic receptor expression, microglial recruitment assays, measurement of LAMP2 expression, evaluation of autophagy-lysosomal function in astrocytes
Upregulated phagocytic receptors, increased microglial Aβ phagocytosis, enhanced astrocytic autophagy-lysosomal function
Increased phagocytic receptor expression, enhanced microglial recruitment and Aβ uptake, elevated LAMP2 expression and improved autophagy-lysosomal function
