Intravitreal injections of IGF-I were administered to rd10 mice to evaluate neuroprotective effects on photoreceptor degeneration in vivo. The experiment assessed photoreceptor cell death using TUNEL staining in retinal sections and quantified free nucleosomes in retinal extracts by ELISA. Microglial cell number and distribution were analyzed using immunolabeling with Cd11b and Iba1 antibodies. The study demonstrated that in vivo IGF-I treatment significantly reduced the number of TUNEL-positive nuclei in rd10 retinas, confirming the neuroprotective effects observed in the ex vivo experiments.
