Apoptosis-Inducing Factor (AIF) is a crucial flavoprotein that plays dual roles in both normal mitochondrial function and programmed cell death. As a key mediator of caspase-independent apoptosis, AIF translocates from mitochondria to the nucleus upon apoptotic stimuli, where it triggers large-scale DNA fragmentation and chromatin condensation.
Structure
The AIF protein contains several distinct structural domains:
- N-terminal mitochondrial targeting sequence (MTS): First 50 amino acids form an amphipathic helix that directs mitochondrial import
- FAD-binding domain: Comprises residues 150-400 and binds FAD cofactor essential for NADH oxidase activity

Role in Disease
Charcot-Marie-Tooth Disease Type 4 (CMTX4)
Recessive mutations in AIFM1 cause X-linked Charcot-Marie-Tooth disease type 4, characterized by peripheral neuropathy, axonal degeneration, and sometimes cognitive impairment<sup>[2]</sup>.
Combined Oxidative Phosphorylation Deficiency (COXPD)
Loss-of-function mutations lead to severe multisystem disorders affecting brain development, causing encephalomyopathy, lactic acidosis, and early-onset neurodegeneration<sup>[3]</sup>.
Alzheimer's Disease
