Overview DRD1 (Dopamine Receptor D1) is the prototypical D1-like dopamine receptor and a critical mediator of dopaminergic signaling in the brain[@dopamine2018][@dopamine2020]. It is a G protein-coupled receptor (GPCR) that activates adenylate cyclase, increasing intracellular cAMP levels upon dopamine binding. DRD1 is highly expressed in the striatum, prefrontal [cortex](/brain-regions/cortex), and other brain regions involved in motor control, reward, and cognition[@drd2021]. Dysregulation of DRD1 signaling is implicated in Parkinson's disease, schizophrenia, and other neurological disorders.
Structure DRD1 has characteristic GPCR features:
Seven transmembrane domains : Form the ligand-binding pocket
Extracellular N-terminus : Contains glycosylation sites
Intracellular C-terminus : Contains phosphorylation sites
Third intracellular loop : Couples to G proteins
Dimerization interface : Can form functional dimers
The receptor adopts the canonical GPCR fold with an orthosteric binding site accessible from the extracellular side[@structure2019].
