Introduction
ROBO3 (Roundabout Guidance Receptor 3) is a transmembrane receptor protein that plays a critical role in axon guidance and neuronal development. Unlike other ROBO family members, ROBO3 has a unique function in promoting midline crossing of axons rather than repelling them. ROBO3 is essential for the development of commissural [neurons](/entities/neurons) in the spinal cord and brainstem. Mutations in ROBO3 cause Horizontal Gaze Palsy with Progressive Scoliosis (HGPPS), a congenital disorder characterized by abnormal eye movements and scoliosis. Recent research suggests that ROBO3 and the Slit-Robo pathway may also play roles in adult brain function and could be implicated in neurodegenerative diseases. [@robo2010]

Structure
ROBO3 is a large type I transmembrane protein with the following domain architecture: [@slitrobo2015]
- Extracellular domain:
- 5 Immunoglobulin (Ig) domains: Mediate ligand binding and homophilic/heterophilic interactions
- 3 Fibronectin type III (FNIII) domains: Support protein-protein interactions
- Multiple conserved cysteine residues for domain stability
- Transmembrane domain: Single pass α-helical membrane-spanning region
- Cytoplasmic domain:
- Multiple conserved motifs for signaling
- Contains conserved CC0, CC1, CC2, CC3 motifs
- Proline-rich regions for SH3 domain interactions
- Potential phosphorylation sites
- Lacks the conserved ROBO intracellular inhibitory domain found in ROBO1/2
- Has distinct signaling properties that promote axon crossing
Normal Function
Axon Guidance
ROBO3 plays a unique role in neuronal circuit formation: [@axon2019]
Signaling Mechanisms
ROBO3 signals through multiple intracellular pathways: [@midline2012]
- Rho GTPase regulation: Modulates Rac, Rho, Cdc42 activity
- Akt pathway: PI3K/Akt signaling downstream of ROBO3
- MAPK/ERK pathway: Regulates gene expression
- Pak1 activation: p21-activated kinase signaling
Expression Pattern
ROBO3 is expressed in: [@robo2014]
- Developing spinal cord (commissural neurons)
- Brainstem (pons, medulla)
- [Cerebellum](/brain-regions/cerebellum)
- Corpus callosum
- Specific neuronal populations in the adult brain
Role in Neurodegeneration
Alzheimer's Disease
Emerging evidence links ROBO3 to AD: [@robo2016]
Parkinson's Disease
ROBO3 may play roles in PD:
Horizontal Gaze Palsy with Progressive Scoliosis (HGPPS)
ROBO3 mutations cause HGPPS:
- Genetic basis: Autosomal recessive mutations in ROBO3 gene
- Clinical features:
- Absent horizontal eye movements
- Progressive scoliosis
- Cranial nerve abnormalities
- Neuroimaging: Malformed brainstem, abnormal axon crossing
Other Neurological Conditions
- Schizophrenia: Some studies link ROBO3 variants to schizophrenia risk
- Autism spectrum disorders: ROBO3 may contribute to midline development relevant to ASD
- Multiple sclerosis: Possible role in axonal guidance during remyelination
