Overview

The cellular prion protein (PrP<sup>C</sup>) is a GPI-anchored glycoprotein expressed predominantly in the central nervous system[@riek1996]. While its physiological function remains incompletely understood, PrP<sup>C</sup> is best known for its central role in prion diseases—fatal neurodegenerative disorders caused by conformational conversion to the disease-associated isoform PrP<sup>Sc</sup> (scrapie prion protein)[@prusiner2013]. Structure

The prion protein is a 253-amino acid GPI-anchored protein with a distinctive domain organization[@riek1996]:
