This nucleus, containing cholinergic preganglionic neurons, plays an essential role in regulating pupillary responses through its projections to the ciliary ganglion via the oculomotor nerve (cranial nerve III). The cholinergic neurons within this nucleus utilize acetylcholine as their primary neurotransmitter, expressing key cholinergic markers including choline acetyltransferase (ChAT) and the vesicular acetylcholine transporter (VAChT), which are essential for acetylcholine synthesis and packaging into synaptic vesicles (Ehert et al., 2020).
