Introduction
Melanin-Concentrating Hormone (MCH) neurons are a critical population of peptidergic neurons located in the lateral hypothalamus that play essential roles in sleep-wake regulation, energy homeostasis, and metabolic function. First characterized in the 1980s, MCH neurons have emerged as key players in the neurobiology of sleep disorders and have significant implications for understanding neurodegenerative diseases. MCH neurons constitute a relatively small but highly influential population in the hypothalamus. They produce and release the neuropeptide melanin-concentrating hormone, which acts on two G protein-coupled receptors (MCHR1 and MCHR2) distributed throughout the brain. These neurons are distinct from orexin/hypocretin neurons, which are located in the same region but have opposing functions—while orexin neurons promote wakefulness, MCH neurons promote sleep.
