Overview

CCAAT/Enhancer Binding Protein Delta (CEBPD) is a transcription factor belonging to the CCAAT/Enhancer Binding Protein (C/EBP) family. This family of basic leucine zipper (bZIP) transcription factors is involved in the regulation of genes involved in inflammation, immune response, metabolism, cell differentiation, and proliferation.
Function
Normal Function
The C/EBP family proteins are transcription factors that bind to CCAAT motifs in gene promoters and enhancers. They function as homo- or heterodimers and regulate gene expression through their leucine zipper DNA-binding domains.
CEBPD is expressed in various tissues including brain, where it plays important roles in:
- Glial Cell Function: Regulation of astrocyte and microglial activation
- Inflammatory Response: Control of pro-inflammatory cytokine expression
- Metabolic Regulation: Modulation of glucose and lipid metabolism
- Cell Differentiation: Involvement in cellular differentiation programs
Expression Pattern
In the brain, CEBPD is primarily expressed in:
- [Astrocytes](/entities/astrocytes) (particularly in gray matter)
- Microglial cells
- Certain neuronal populations
- Oligodendrocyte precursor cells
Expression is often upregulated in response to inflammatory stimuli, injury, or disease states.
Disease Associations
CEBPD has been implicated in several neurodegenerative and inflammatory diseases:
| Disease | Role | Evidence |
|---------|------|----------|
| [Alzheimer's Disease](/diseases/alzheimers-disease) | Neuroinflammation | Upregulated in AD brain; regulates inflammatory cytokines |
| [Parkinson's Disease](/diseases/parkinsons-disease) | Glial activation | Altered expression in PD models; contributes to neuroinflammation |
| Multiple Sclerosis | Demyelination | Regulates immune cell function; implicated in MS pathogenesis |
| Amyotrophic Lateral Sclerosis | Neuroinflammation | Altered in ALS models and patient tissue |
The involvement in neurodegeneration is primarily related to the regulation of inflammatory processes and glial cell function.
Therapeutic Implications
Targeting CEBPD signaling pathways may offer therapeutic strategies for neurodegenerative diseases:
Anti-inflammatory Approaches: Modulating C/EBP activity could reduce harmful neuroinflammation
Gene Therapy: Selective modulation of C/EBP expression in glia
Small Molecule Inhibitors: Development of compounds targeting C/EBP-DNA interactions
Combination Therapies: Targeting C/EBP alongside other therapeutic approachesKey Publications
