Overview

CERNUNNOS (also known as X-linked childhood cerebellar ataxia) is a DNA repair protein that plays a critical role in the non-homologous end joining (NHEJ) pathway of DNA double-strand break repair. The protein is essential for maintaining genomic stability in neuronal cells, which are particularly vulnerable to DNA damage due to their post-mitotic nature and high metabolic activity. CERNUNNOS interacts with the Ku70/Ku80 heterodimer and DNA-PKcs to facilitate the repair of DNA double-strand breaks, a process crucial for neuronal survival and function [1](https://pubmed.ncbi.nlm.nih.gov/19483685/).

Molecular Function

CERNUNNOS functions as a scaffold protein in the NHEJ repair complex, facilitating the recruitment and assembly of key repair proteins at sites of DNA damage. The protein contains multiple tetratricopeptide repeat (TPR) domains that mediate protein-protein interactions with molecular chaperones and co-chaperones, including Hsp90 and various co-chaperones of the TPR family [2](https://pubmed.ncbi.nlm.nih.gov/19011601/).
