DUSP1 (Dual Specificity Phosphatase 1), also known as MAPK Phosphatase-1 (MKP-1), is an inducible MAP kinase phosphatase that dephosphorylates both tyrosine and threonine residues on MAP kinases [1](https://doi.org/10.1016/S0960-9822(01)00156-3). It is a key negative regulator of stress-activated protein kinase signaling pathways.
Structure
DUSP1 is a 367 amino acid protein with:
N-terminal non-catalytic domain: Contains binding sites for MAP kinases and regulatory proteins
C-terminal catalytic domain: DSP domain with dual-specificity phosphatase activity
Kinase interaction motif (KIM): Mediates binding to MAP kinases
Nuclear localization signal (NLS): Regulates subcellular localization
The protein adopts a fold typical of PTP family phosphatases, with a conserved HCX5R motif in the active site [2](https://doi.org/10.1074/jbc.275.10.7466).
