General Transcription Factor IIH Subunit 2 (GTF2H2) is a 44 kDa protein that forms an essential core component of the TFIIH transcription factor complex, also known as the general transcription factor TFIIH. The TFIIH complex is a multi-subunit molecular machine (~500 kDa total) that plays a dual critical role in eukaryotic cells: it opens DNA during transcription initiation by RNA polymerase II (RNAPII) and it executes nucleotide excision repair (NER) pathway to remove bulky DNA lesions caused by UV radiation, oxidative damage, and chemical adducts[@egly2011]. GTF2H2 participates directly in both of these fundamental cellular processes through its structural and enzymatic roles within the complex.
