Introduction

The p75 neurotrophin receptor (p75NTR), encoded by the [NGFR](/genes/ngfr) gene, is a member of the tumor necrosis factor (TNF) receptor superfamily that functions as a key regulator of neuronal survival, death, and differentiation [1](https://pubmed.ncbi.nlm.nih.gov/12444493/). Originally discovered as the nerve growth factor (NGF) receptor, p75NTR has emerged as a critical player in neurodegenerative diseases, including Alzheimer's disease (AD), Parkinson's disease (PD), and amyotrophic lateral sclerosis (ALS).[@costantini2005] Unlike Trk receptor tyrosine kinases that mediate pro-survival signaling, p75NTR can activate both pro-survival and pro-apoptotic pathways depending on cellular context, co-receptor expression, and ligand availability.

p75NTR exhibits a unique ability to bind all neurotrophins (NGF, BDNF, NT-3, NT-4) with relatively low affinity, serving as a molecular switch that determines cellular outcomes in response to neurotrophin signaling. This receptor plays essential roles in development, synaptic plasticity, and the pathogenesis of multiple neurodegenerative disorders [2](https://pubmed.ncbi.nlm.nih.gov/11860281/).
