Overview
ROCK1 is a protein involved in cytoskeleton regulation, cell motility, and Rho kinase signaling. This protein plays a role in neurodegenerative diseases and represents a potential therapeutic target.
Structure
ROCK1 is a serine/threonine kinase with a modular structure:
- Rho-binding domain (RBD): At N-terminus, binds active RhoA
- Coiled-coil region: Mediates dimerization and localization
- Kinase domain: At C-terminus, catalyzes phosphorylation of substrates
- PH domain: Regulates activity and localization
ROCK1 exists in an autoinhibited conformation that is relieved by RhoA binding or caspase cleavage.
Normal Function
ROCK1 regulates numerous cellular processes:
- Actin cytoskeleton: Phosphorylates myosin light chain (MLC), LIM kinase
- Cell adhesion: Modulates focal adhesion dynamics
- Neuronal development: Axon guidance, dendritic spine formation
- Synaptic plasticity: Regulates AMPA receptor trafficking
- Smooth muscle contraction: Controls actomyosin contractility
In the brain, ROCK1 regulates neuronal morphology, migration, and synaptic function.
Role in Disease
ROCK1 dysregulation contributes to neurodegenerative diseases:
- Alzheimer's Disease: Enhances [tau](/proteins/tau) phosphorylation; promotes [amyloid-beta](/proteins/amyloid-beta) toxicity
- Parkinson's Disease: Facilitates [alpha-synuclein](/proteins/alpha-synuclein) phosphorylation; affects dopaminergic neuron survival
- Stroke: Increases [blood-brain barrier](/entities/blood-brain-barrier) permeability; promotes excitotoxicity
- ALS: Contributes to cytoskeletal defects and axonal transport impairment
Therapeutic Targeting
- ROCK inhibitors: Y-27632, fasudil, aspirin
- RhoA inhibitors: Targeting upstream activation
- Anti-inflammatory agents: Reducing downstream effects
