Overview
NEK7 (NIMA-related kinase 7) is a serine/threonine kinase belonging to the NEK (Never in Mitosis A-related kinase) family. NEK7 is a 34 kDa serine/threonine kinase that plays dual roles in cell biology:
Mitotic Functions
NEK7 participates in mitotic spindle assembly and cytokinesis. It is activated by [NEK9](/genes/nek9) during mitosis and phosphorylates downstream targets required for proper centrosome separation and bipolar spindle formation. NEK7 and the closely related [NEK6](/genes/nek6) share approximately 87% sequence identity in their kinase domains and have partially overlapping roles in cell division.
NLRP3 Inflammasome Activation
The most disease-relevant function of NEK7 is its role as an essential upstream mediator of [NLRP3](/genes/nlrp3) inflammasome assembly. NEK7 directly binds the leucine-rich repeat (LRR) domain and nucleotide-binding domain (NBD) of NLRP3 through its C-terminal kinase domain. This interaction is required for NLRP3 oligomerization, [ASC](/genes/pycard) speck formation, and subsequent [caspase-1](/genes/casp1) activation.
Key mechanistic details include:
- NEK7 acts downstream of potassium efflux, a common signal triggered by diverse NLRP3 stimuli (ATP, nigericin, particulate matter, mitochondrial reactive oxygen species)
- The NEK7-NLRP3 interaction is cell cycle-dependent: during interphase NEK7 is available for NLRP3 binding, but during mitosis it is sequestered for spindle functions — explaining why inflammasome activation is suppressed in dividing cells
- NEK7 kinase activity is dispensable for NLRP3 activation; the interaction is structural rather than catalytic

Other Immune Functions
