== Function == NRCAM (Neuronal Cell Adhesion Molecule) is a member of the L1 family of cell adhesion molecules (L1-CAMs). It is a transmembrane glycoprotein with six Ig-like domains and five fibronectin type III repeats in its extracellular region. NRCAM mediates homophilic (NRCAM-NRCAM) and heterophilic (NRCAM-ankyrin-G) interactions that are essential for neuronal migration, axon guidance, and synapse formation. [@sakurai2011]

NRCAM plays critical roles in: [@maness2004]
Neuronal Migration: During cortical development, NRCAM guides neuronal precursors to their proper positions
Axon Guidance: Functions as a guidance molecule for developing axons, particularly in the optic chiasm and corticospinal tract
Synapse Formation: Localizes to synaptic membranes and participates in synapse assembly and function
Node of Ranvier Formation: Clusters at nodes of Ranvier in myelinated axons alongside other L1-CAMs (L1, Nr-CAM)
== Disease Associations == [@sakurai2011a] NRCAM variants have been associated with: [@pollerberg2013]
Autism Spectrum Disorder (ASD): Multiple studies have identified NRCAM variants in individuals with autism
Multiple Sclerosis (MS): Gene associations reported in GWAS studies
Epilepsy: Rare variants may contribute to seizure susceptibility
Intellectual Disability: Some variants associated with neurodevelopmental disorders
== Expression == [@hansen2014] NRCAM shows high expression in: [@matsumoto2008]

NRCAM (Neuronal Cell Adhesion Molecule) encodes a neuronal adhesion molecule of the L1 family that mediates cell-cell adhesion and axon guidance. NRCAM plays important roles in nervous system development, including neurite outgrowth, myelination, and synapse formation. NRCAM is expressed in the developing brain and continues to be expressed in adult neurons. Genetic variants in NRCAM have been associated with autism spectrum disorder and multiple sclerosis.
