Pathway Diagram
AXIN2 (Axis Inhibition Protein 2), also known as Conductin or Axil, is a critical negative regulator of the Wnt/beta-catenin signaling pathway. Like its homolog AXIN1, AXIN2 serves as a scaffold protein in the beta-catenin destruction complex, facilitating the phosphorylation and degradation of beta-catenin. However, AXIN2 has distinct expression patterns and functions in the adult brain, particularly in neural stem cells, synaptic plasticity, and stress responses. Dysregulated AXIN2 expression has been implicated in [Alzheimer's disease](/diseases/alzheimers-disease), [Parkinson's disease](/diseases/parkinsons-disease-disease), and cancer.

Overview
AXIN2 encodes a protein of 778 amino acids that shares significant homology with AXIN1, particularly in the DIX domain, RGS domain, and AXIN domain. However, AXIN2 has unique functions in adult tissues, particularly in stem cell populations and the nervous system.
Unlike AXIN1, AXIN2 expression is inducible by Wnt signaling itself, creating a negative feedback loop that limits the duration and intensity of Wnt pathway activation. This feedback mechanism is crucial for maintaining proper Wnt signaling levels during development and in adult tissues[@lustig2002].
Molecular Structure
AXIN2 contains similar domains to AXIN1:
- DIX domain: Mediates oligomerization and Dishevelled interaction
- RGS domain: Regulatory protein-protein interactions
- AXIN domain: APC and homodimer binding
- C-terminal domain: Beta-catenin and [GSK-3β](/entities/gsk3-beta) binding
Normal Function in the Nervous System
Neural Stem Cells
AXIN2 is expressed in neural stem cells (NSCs) in the adult subventricular zone (SVZ) and dentate gyrus subgranular zone (SGZ):
- Stem cell maintenance: AXIN2+ NSCs represent the active stem cell population in adult neurogenic niches[@garcaverdugo2011].
- Wnt feedback: AXIN2 expression in NSCs provides negative feedback to regulate Wnt-driven proliferation.
- Neurogenesis: AXIN2 regulates neuronal differentiation from adult NSCs.
Synaptic Function
In mature [neurons](/entities/neurons), AXIN2 localizes to synapses and participates in:
- Synaptic plasticity: AXIN2 regulates both [LTP](/mechanisms/long-term-potentiation) and LTD, similar to AXIN1[@zhang2014].
- Dendritic spine dynamics: AXIN2 controls spine density and morphology through Wnt-independent mechanisms.
- Synaptic protein turnover: AXIN2 regulates the degradation of synaptic proteins via the proteasome.
Stress Response
AXIN2 is involved in cellular stress responses:
- DNA damage response: AXIN2 interacts with p53 and regulates [apoptosis](/entities/apoptosis) under genotoxic stress[@song2007].
- Oxidative stress: AXIN2 expression is modulated by oxidative stress and may protect neurons.
- ER stress: AXIN2 participates in the [unfolded protein response](/entities/unfolded-protein-response) (UPR).

Parkinson's Disease
AXIN2 and Neuropsychiatric Disorders
Beyond neurodegenerative diseases, AXIN2 has been linked to neuropsychiatric conditions:
AXIN2 in Neural Progenitor Fate
AXIN2 plays a critical role in fate decisions:
Expression Pattern
AXIN2 is expressed in:
- Adult neurogenic niches: Subventricular zone, dentate gyrus subgranular zone
- [Hippocampus](/brain-regions/hippocampus): Primarily in dentate gyrus granule cells
- Cerebral [cortex](/brain-regions/cortex): Lower expression than AXIN1
- Neural progenitors: During development and in adult stem cells
