Endonuclease G (ENDOG) is a mitochondrial nuclease enzyme encoded by the ENDOG gene located on chromosome 14q11.2. Under normal conditions, the protein maintains a housekeeping role in mitochondrial DNA homeostasis, participating in the degradation of damaged or oxidatively modified mitochondrial DNA. This function helps preserve mitochondrial genome integrity and prevents the accumulation of mutagenic lesions. ENDOG possesses non-specific endonucleolytic activity, capable of cleaving double-stranded DNA in a magnesium-dependent manner, generating characteristic internucleosomal fragments of approximately 50-300 kilobases.
