Function
KIF13A is a member of the kinesin-3 family that participates in endocytic trafficking and synaptic vesicle transport. It localizes to growth cones and [dendritic spines](/mechanisms/dendritic-spines), where it regulates spine morphology and synaptic plasticity. KIF13A is involved in the transport of [amyloid precursor protein](/entities/app-protein) (APP) and is implicated in Alzheimer's disease pathogenesis.
Axonal Transport in Neurodegeneration
Axonal transport is essential for maintaining neuronal health. Defects in kinesin-mediated transport are implicated in:
- Hereditary Spastic Paraplegia (HSP): Mutations in KIF1A, KIF5A cause HSP
- Charcot-Marie-Tooth Disease: KIF1B, KIF1A, KIF5A mutations
- Alzheimer's Disease: Impaired transport contributes to amyloid and [tau](/proteins/tau) pathology
- Parkinson's Disease: Transport defects affect [alpha-synuclein](/proteins/alpha-synuclein) clearance
Disease Associations
- Hereditary Spastic Paraplegia: KIF1A (SPG30), KIF5A (SPG10)
- Charcot-Marie-Tooth Disease: KIF1B (CMT2A), KIF1A, KIF5A
- Intellectual Disability: KIF1B, KIF5C mutations
- Parkinson's Disease: KIF1A, KIF13A involvement
Expression
KIF13A is predominantly expressed in:
- [Neurons](/entities/neurons) (both central and peripheral)
- Highest in cerebral [cortex](/brain-regions/cortex), [hippocampus](/brain-regions/hippocampus), cerebellum
- Localizes to axons and dendrites
Therapeutic Implications
- Enhancing axonal transport as a therapeutic strategy
- Gene therapy approaches for HSP
- Small molecules to activate kinesin motors
