Structure PLIN2 (also known as adipophilin) is a member of the perilipin family of proteins that regulate lipid droplet dynamics[@kimmel2010]. The protein contains:
N-terminal PAT domain (~100 aa) involved in lipid droplet targeting
Central hydrophobic regions for membrane association
C-terminal region mediating protein-protein interactions
Conserved 11-mer repeats involved in fatty acid binding[@bickerton2017]
Normal Function PLIN2 is a key regulator of cellular lipid metabolism:
Coats the surface of cytoplasmic lipid droplets
Protects triglycerides from lipolysis by inhibiting hormone-sensitive lipase
Facilitates fatty acid uptake and storage
Regulates lipid droplet trafficking between cellular compartments
Involved in [ER](/mechanisms/endoplasmic-reticulum-stress) lipid droplet biogenesis[@walther2012]
Role in Neurodegeneration
Alzheimer's Disease PLIN2 is strongly implicated in [Alzheimer's disease](/diseases/alzheimers-disease) pathogenesis:
Elevated in AD brain : PLIN2 expression is significantly increased in AD [hippocampus](/brain-regions/hippocampus) and prefrontal [cortex](/brain-regions/cortex)[@lerch2015]
Lipid droplet accumulation : AD [neurons](/entities/neurons) show increased lipid droplet accumulation, with PLIN2 as a major component
[Aβ](/proteins/amyloid-beta) interaction : [Amyloid-beta](/proteins/amyloid-beta-protein) promotes PLIN2 upregulation through [PPARγ](/entities/pparg) signaling
[Ferroptosis](/entities/ferroptosis) connection : PLIN2-associated lipid droplets may modulate [ferroptosis](/mechanisms/ferroptosis-pathway) in AD[@zhang2022]
Lipid metabolism dysregulation : Altered PLIN2 reflects broader lipid metabolism disturbances in AD[@han2021]
Parkinson's Disease
Increased PLIN2 observed in [Parkinson's disease](/diseases/parkinsons-disease) substantia nigra dopaminergic neurons[@garciagarcia2013]
Lipid droplet accumulation linked to [alpha-synuclein](/proteins/alpha-synuclein) aggregation
May reflect impaired [autophagy](/mechanisms/autophagy-lysosomal-pathway)-lipophagy pathway
Amyotrophic Lateral Sclerosis
PLIN2 upregulation in [ALS](/diseases/amyotrophic-lateral-sclerosis) motor neurons
Lipid droplet accumulation precedes motor neuron degeneration in some models
Linked to altered lipid metabolism in [C9orf72](/genes/c9orf72)-related ALS[@liu2017]
Huntington's Disease
Elevated PLIN2 in [Huntington's disease](/diseases/huntington-disease) striatal neurons[@block2020]
May contribute to metabolic dysfunction through lipid droplet dysregulation
Therapeutic Targeting
PPAR Modulators : [PPARγ](/entities/pparg) agonists may normalize PLIN2 expression
Lipid Metabolism Modulators : Targeting fatty acid oxidation pathways[@chandra2022]
[Autophagy](/entities/autophagy) Enhancers : [Rapamycin](/therapeutics/rapamycin-tauopathy) and related compounds to clear lipid droplets
Dietary Interventions : Omega-3 fatty acid supplementation may modulate PLIN2[@dyall2017]
