Overview
TYROBP (TYRO Protein Tyrosine Kinase-Binding Protein), commonly known as DAP12 (DNAX-Activation Protein 12), is a transmembrane signaling adaptor protein that plays critical roles in innate immunity and microglial function in the central nervous system. DAP12 partners with multiple immunoreceptors, most notably [TREM2](/proteins/trem2), to transduce activation signals that regulate microglial survival, phagocytosis, and inflammatory responses[@lanier1998].
Mutations in TYROBP cause Nasu-Hakola disease (polycystic lipomembranous osteodysplasia with sclerosing leukoencephalopathy, PLOSL), a rare autosomal recessive disorder characterized by presenile dementia and bone cysts. Additionally, DAP12 signaling is increasingly recognized as a key pathway in Alzheimer's disease, Parkinson's disease, and other neurodegenerative conditions[@paloneva2000][@colonna2003].
Structure and Domain Architecture
TYROBP is a small 113-amino acid type I transmembrane protein:
- Very short extracellular region
- Contains cysteine residues for disulfide linkage to partner receptors
- Mediates non-covalent association with receptor partners
- Single α-helix with a critical aspartic acid residue (Asp50)
- Asp50 forms a salt bridge with a lysine in the partner receptor's transmembrane domain
- This electrostatic interaction drives receptor-adaptor pairing[@feng2010]
- Contains the ITAM (Immunoreceptor Tyrosine-based Activation Motif)
- ITAM sequence: YxxL/I...YxxL/I (Tyr71 and Tyr86)
- Phosphorylation sites that recruit SYK and ZAP70 kinases
