Overview

SIN3A (SWI-Independent 3A) is a ~145 kDa scaffold protein that serves as the core component of the SIN3-HDAC (histone deacetylase) transcriptional repression complex. Originally discovered in yeast as a global transcriptional repressor, SIN3A has evolved into a central regulator of gene expression programs critical for neuronal development, synaptic plasticity, DNA repair, and cellular homeostasis [@silverstein2010]. Through its ability to recruit HDAC1 and HDAC2 to specific genomic loci, SIN3A modulates chromatin acetylation states to silence target genes. This modular architecture enables precise targeting of repression to specific genomic sites through interaction with DNA-bound transcription factors and chromatin regulators. In neurons, SIN3A-regulated genes include those involved in synaptic function, neuronal survival, and stress responses.
