Introduction
NCOA1 (Nuclear Receptor Coactivator 1), also known as SRC-1 (Steroid Receptor Coactivator-1), is a member of the p160 family of nuclear receptor coactivators[@ncoa2000]. It plays a critical role in modulating gene expression by enhancing the transcriptional activity of nuclear receptors and other transcription factors. NCOA1 is widely expressed in the brain and has been implicated in various neurological processes including neuroprotection, synaptic plasticity, and neurodegenerative disease pathogenesis[@nuclear2012].
Molecular Function
Structure
NCOA1 contains several functional domains:
- N-terminal basic helix-loop-helix (bHLH) domain: DNA binding and dimerization
- PAS domains (PAS-A, PAS-B): Protein-protein interactions and sensing environmental signals
- C-terminal activation domains (AD1, AD2): Interaction with chromatin remodeling complexes and basal transcription machinery[@structural2002]
Mechanism of Action
NCOA1 functions as a molecular bridge:
Role in the Brain
Neurodevelopment
During brain development, NCOA1 regulates:
- Neuronal differentiation and migration
- Astrocyte and oligodendrocyte development
- Synapse formation and maturation
- Axonal guidance and myelination[@ncoa2011]
Adult Brain Function
In the mature brain, NCOA1 modulates:
