Peripeduncular Nucleus (PPN) Neurons

Introduction

The peripeduncular nucleus (PPN), also known as the peripeduncular nucleus of the ventral thalamus or the posterior intralaminar nucleus, is a midline thalamic structure that occupies a strategic position between the medial geniculate body, the inferior colliculus, and the cerebral peduncle. First described in early neuroanatomical studies, the PPN has emerged as a critical node in circuits processing pain, autonomic information, and sleep-wake transitions. Its unique connectivity pattern, receiving input from spinal and brainstem pain pathways while projecting to frontal cortical and limbic structures, positions it as a key relay for integrating somatosensory information with higher-order cognitive and emotional processing. Interest in the PPN has grown considerably in recent years due to its involvement in neurodegenerative diseases. Both Alzheimer's disease and Parkinson's disease are associated with pathological changes in the PPN, and these alterations may contribute to the characteristic symptoms of these disorders, including pain hypersensitivity, autonomic dysfunction, and sleep disturbances.
