Overview

Leucine-rich repeat kinase 2 (LRRK2), also known as dardarin, is a large multi-domain protein encoded by the LRRK2 gene (located at chromosome 12q12) that has emerged as one of the most important genetic contributors to Parkinson's disease (PD). Discovered in 2004 through linkage analysis of the "S TASK" family, LRRK2 mutations are now recognized as the most common cause of genetically determined PD, accounting for approximately 5-10% of familial cases and 1-5% of sporadic cases worldwide[@paisan-ruiz2019].
