Overview
LRP8 (Low-Density Lipoprotein Receptor-Related Protein 8), also known as ApoER2, is a member of the LDL receptor family primarily expressed in the brain. It serves as a critical receptor for the extracellular matrix protein Reelin, playing essential roles in neuronal migration during development, synaptic plasticity, and cognitive function. LRP8 also functions as a receptor for [APOE](/proteins/apoe-protein)-containing lipoproteins, linking lipid metabolism to neuronal function. Genetic variants in LRP8 have been associated with Alzheimer's disease risk, stroke, and neurodevelopmental disorders.[@supsup2006]
Gene Structure
The LRP8 gene:
- Located on chromosome 1p31.3
- Contains 31 exons spanning approximately 40 kb
- Produces multiple splice variants with tissue-specific expression
- Alternative splicing generates isoforms with differing cytoplasmic tails
Splice Variants
| Isoform | Expression | Key Features |
|---------|------------|--------------|
| Full-length | Brain | Contains NPXY motifs for endocytosis |
| Variant 1 | Testis | Lacks part of cytoplasmic tail |
| Variant 2 | Various | Alternative exon usage |
Protein Structure
LRP8 is a modular receptor protein:
Extracellular Domain (1-726)
Transmembrane Domain (727-749)
- Single-pass transmembrane helix
Cytoplasmic Tail (750-870)
- NPxY motifs: Two motifs for endocytosis and signaling
- Phosphorylation sites: Regulate protein interactions
Normal Function
Reelin Signaling
LRP8 is a critical receptor for Reelin:[@supsup2014]
