Neurogranin (Ng)

Neurogranin (Ng) is a postsynaptic neuronal protein that serves as a sensitive biomarker for synaptic dysfunction in Alzheimer's disease and other neurodegenerative disorders[@portelius2015].

Overview

Neurogranin is a 78-amino acid protein encoded by the RC3 (RC3/Ng) gene, also known as neurogranin or RC3. It is highly enriched in dendritic spines of excitatory neurons in the hippocampus and cerebral cortex[@dazmoreno2010]. As a biomarker, neurogranin reflects the loss of synaptic integrity, which is a hallmark of cognitive decline in neurodegenerative diseases.

Normal Physiological Function

Neurogranin plays several critical roles in synaptic plasticity[@derkach2007]:

  • Calmodulin Binding: Ng binds to calmodulin in the absence of calcium, regulating calmodulin availability for downstream signaling
  • Synaptic Plasticity: Involved in long-term potentiation (LTP) and memory formation
  • PKC Substrate: Major substrate for protein kinase C (PKC) phosphorylation
  • Dendritic Spine Morphogenesis: Regulates the structure and function of dendritic spines
  • Role as a Biomarker
