Overview

Synaptic vesicles undergo a coordinated cycle of trafficking, docking, priming, fusion, release, and recycling to sustain neurotransmission. This cycle involves over 100 coordinated in tightly regulated steps. The synaptic vesicle cycle is fundamental to neuronal communication. Each synaptic vesicle must be loaded with neurotransmitters, transported to the active zone, undergo molecular priming steps, fuse with the presynaptic membrane in response to calcium influx, release its contents, and then be recycled through endocytosis for reuse. This process consumes substantial ATP and requires precise protein-protein interactions that are vulnerable to disruption by pathological implicated in neurodegenerative [@de2008][@rizzoli2014].
