GBA Pathway in Parkinson's Disease

Overview

The GBA gene encodes glucocerebrosidase (GCase), a lysosomal hydrolase essential for glycolipid metabolism. Heterozygous mutations in GBA represent the most significant genetic risk factor for Parkinson's disease (PD) identified to date, increasing PD risk by approximately 5- to 7-fold. The GBA-PD association represents a crucial link between lysosomal storage disorders and neurodegenerative disease, revealing fundamental mechanisms of protein aggregation and cellular vulnerability that transcend traditional disease boundaries.
