Overview
This experiment addresses a critical therapeutic gap in [Parkinson's disease](/diseases/parkinsons-disease) (PD): the role of lipid droplet accumulation and lysosomal dysfunction in dopaminergic neuron degeneration. While the [autophagy](/entities/autophagy)-lysosome pathway has been targeted in previous experiments, the specific contribution of lipid droplet metabolism and lipophagy remains underexplored.
This two-phase preclinical experiment will systematically test pharmacological interventions targeting the lipid droplet-lysosome axis in PD models, progressing from in vitro screening to in vivo validation.
Hypothesis
Primary Hypothesis: Pharmacological modulation of lipid droplet metabolism and lysosomal function will reduce α-synuclein pathology and preserve dopaminergic neuron viability in PD models, with lipophagy activators showing superior efficacy compared to lipid droplet dispersers.

Aim 1: In Vitro Screening in iPSC-Derived Dopaminergic Neurons
- Screen therapeutic compounds targeting lipid droplet-lysosome axis
- Use iPSC-derived midbrain dopaminergic [neurons](/entities/neurons) from PD patients
- Identify lead compounds based on efficacy and safety profile
Aim 2: In Vivo Validation in AAV-α-Synuclein Mouse Model
- Validate top compounds in established α-synuclein propagation model
- Assess motor behavior, neuropathology, and mechanism of action
- Establish dose-response relationships and PK/PD profiles

iPSC-Derived Dopaminergic Neuron Platform
Cell Lines:
- 3 patient-derived iPSC lines with known PD mutations:
- [LRRK2](/entities/lrrk2) G2019S (n=2 lines)
- [GBA1](/entities/gba) N370S carrier (n=1 line)
- 2 isogenic controls (CRISPR-corrected)
- Floor plate patterning followed by midbrain floor plate induction
- Day 0-7: Dual SMAD inhibition (SB431542, LDN193189)
- Day 7-14: SHH activation (Purmorphamine, SAG)
- Day 14-28: Floor plate maturation (CHIR99021, FGF8)
- Day 28-45: Dopaminergic neuron maturation
- Quality criteria: >70% TH+ neurons, >50% FOXA2+/Lmx1a+ progenitors
Screening Protocol
Compound Library:
- 9 compounds (3 per category) at 5 concentrations
- Positive controls: Rapamycin (100 nM), Bafilomycin A1 (50 nM)
- Negative controls: DMSO (0.1%)
- 96-well plates, 15,000 neurons/well
- Treatment duration: 7 days (chronic) and 24 hours (acute)
- Three replicates per condition
